上游原料:锌粉 --> Α-球蛋白 --> N-甲基哌嗪 --> 哌嗪(六水) --> 1-甲醛哌嗪
方法一:以六水哌嗪为原料，经甲基化、水解、亚硝化，还原等步骤得到。（1）甲基化将六水哌嗪溶于甲酸中，吸入反应锅内，升温到90℃，搅拌下滴加甲醛约1h，加毕，于90-95℃保温反应1h，减压蒸去水及过量的原料，得甲基甲酰基哌嗪浓缩液。（2）水解将盐酸和水加到上述浓缩液中，搅拌升温至105℃，回流反应2h。减压蒸馏至无馏出液为止，弃去馏出的酸液，残留液冷至15℃，放置过夜，过滤，滤液即甲基哌嗪盐酸盐溶液。（3）亚硝化将水解步骤得到的滤液在30℃滴加亚硝酸钠溶液，加毕，保温反应30min，得甲基硝基哌嗪生成液。（4）还原在亚硝化生成液中加入冰醋酸，冷却，于30-40℃逐步加入锌粉，加毕，于30-40℃保温反应1.5h，冷至15℃左右，放置过夜，甩滤。将此还原滤液加至预先已配置液碱并冷至10℃以下的碱化锅中，充分冷却后送至萃取锅，分3次加入氯仿进行萃取，合并氯仿萃取液，进行常压蒸馏回收氯仿，至液温为85℃时转入精馏锅中，先常压蒸馏至液温为135℃，再进行减压蒸馏，收集120℃（5.33-8.0kPa）馏分即为1-氨基-4-甲基哌嗪。上述四个步骤的总收率约30%。原料消耗定额：六水哌嗪（95%）1148.1kg/t、甲酸（85%）2266.2kg/t、甲醛（37%）1130.8kg/t、锌粉（90%）1900.2kg/t、冰乙酸3759.4kg/t、浓盐酸4046.5kg/t、亚硝酸钠937.6kg/t。
