方法一:由丁醇与三氯化磷反应而得。美国Hooker化学公司在低于大气压力的条件下使丁醇与三氯化磷反应。液态醇进入反应区，便立即蒸发，吸收放热反应所产生的热量，使反应顺利进行。亚磷酸二丁酯的收率以PCI3计可达92%，并随醇用量的增加而提高。
